Abstract: Detailed, comprehensive and reliable characterization of virus vectors are required to ensure their efficacy and safety. We have been focusing on the development of methods for primary and higher order structure of recombinant adeno associated virus vectors (rAAVs). Following the determination of partial specific volumes of full and empty capsids of AAV, the size distribution analysis of rAAVs was achieved with constituent components by multiwavelength sedimentation velocity analytical ultracentrifugation using OPTIMA AUC, by which we could determine the population of free DNA, intermediate particles, aggregates as well as full and empty particles.
